E.coli is Gram-negative bacteria that have the inner (IM) and outer (OM) membranes ,and the periplasmic space contains the peptidoglycan layer.
The leader sequence signal peptide(PelB) was cleaved at the inner membrane, releasing the mature protein (downstream of PelB)into the periplasmic space . In the periplasmic space, the beta-domain(C-terminal) of the protein(IgAP) inserts into the outer membrane to form a β-barrel structure. After formation of the β-barrel pore, the N-passenger domain(F10scFv) was translocated to the cell surface through the pore.
So we can anchor the F10scFv(antibody) on E.coli 's surface,and catch the influenza A virus.
